Interleukin 1 receptor antagonist and neurodegenerative diseases: the future treatment strategy

نویسندگان

چکیده

Neurodegenerative disorders encompass a range of conditions affecting the central nervous system (CNS) in which alterations neuronal structure and cellular dysfunction lead to progressive deterioration. Activation microglia expression inflammatory cytokine interleukin-1 (IL-1) CNS have become almost synonymous with neuroinflammation. Additionally, relentless activation IL-1 signalling pathway has been linked pathogenesis various disease states, ranging from Alzheimer (AD), Parkinson (PD) amyotrophic lateral sclerosis (ALS) multiple (MS). Moreover, growing body evidence inferred that impeding either pharmacologically or genetically models could reduce neuroinflammation delay progression. This review will therefore aim study role neurodegenerative diseases highlight key aspects warrant IL-1Ra as promising target for developing novel treatment conditions.

Exosomes: Future Perspective in Neurodegenerative Diseases

Neurodegeneration is a progressive and irreversible loss of neuronal cells in specific regions of the brain. Alzheimer Diseases (AD) Parkinson Disease (PD) are the most common forms of neurodegenerative diseases in older people. Exosomes are extracellular nanovesicles that have a key role in physiological processes such as intercellular communication, cell migration, angiogenesis, and anti-tumo...

Interleukin-1 receptor antagonist.

IL-1ra is the first described naturally occurring receptor antagonist of any cytokine or hormone-like molecule. IL-1ra is a member of the IL-1 family by three criteria: amino acid sequence homology of 26 to 30% to IL-1 beta and 19% to IL-1 alpha; similarities in gene structure; and common gene localization to human chromosome 2q14. Two structural variants of IL-1ra exist: sIL-1ra, a secretory m...

Treatment of rheumatoid arthritis with interleukin 1 receptor antagonist.

PURPOSE Interleukin -1 receptor antagonist ( IL-1Ra ) is a new option among biotherapies against rheumatoid arthritis ( RA ). THE AIM of this review is to recall the rationale of use of IL-1Ra and to analyse the results available in the current literature. CURRENT KNOWLEDGE AND KEY POINTS Pathophysiological data of RA give a specific position for IL-1 as a potential target for immunotherapy...

Haplotypes of the interleukin-1 receptor antagonist gene, interleukin-1 receptor antagonist mRNA levels and the risk of myocardial infarction.

BACKGROUND The overall effect of the major pro-inflammatory cytokine interleukin-1 (IL-1) on coagulation and fibrinolysis is prothrombotic. We recently found that haplotype 5 (H5) of the gene (IL1RN) coding for the interleukin-1 receptor antagonist (IL-1Ra), the natural inhibitor of IL-1, is associated with an increased risk of venous thrombosis. It is unclear whether variations in IL1RN affect...

Physiologic role of interleukin-1 receptor antagonist

Recent studies have described the spontaneous development of arthritis or vasculitis in IL-1 receptor antagonist (IL-1Ra) knockout mice bred on specific and different genetic backgrounds. The levels of both secreted and intracellular isoforms of IL-1Ra produced in the rheumatoid joint or in the arterial wall may not be adequate to effectively inhibit the excess amounts of locally produced IL-1....
